An individual microvillus\u00a0is simple in\u00a0structure, consisting of a supporting core of ~25 actin filaments\u00a0bundled\u00a0in parallel by\u00a0villin, fimbrin, and espin. Remarkably, microvilli biogenesis persists in\u00a0mice\u00a0lacking all three of these factors, suggesting the existence of unknown\u00a0bundlers. We identified Mitotic Spindle Positioning (MISP) as an actin binding\u00a0factor\u00a0that localizes specifically to the rootlet end of the microvillus. MISP\u00a0promotes rootlet\u00a0elongation in cells, and purified MISP exhibits potent\u00a0filament bundling activity\u00a0in\u00a0vitro. MISP-bundled filaments also recruit\u00a0fimbrin, which further elongates and\u00a0stabilizes bundles. MISP confinement to\u00a0the rootlet is enforced by ezrin, which\u00a0prevents decoration of the\u00a0membrane-wrapped distal end of the core bundle.
